The currently recommended by WHO for prevention individuals exposed to rabies virus (RV) is pre-exposure prophylaxis (PEP) through the administration of three doses of rabies vaccine by intradermal (ID) or intramuscular (IM) route. The previous study showed that the persistence of protective antibodies titer in the IM schedule is more long-lived, suggesting a different clearance mechanism of antibody. Comparison of in vivo and in vitro transfection of antibodies against rabies MV&Z

This study describes the comparison of in vivo and in vitro transfection of antibodies to use as a new mechanism for antiviral therapy against rabies. The transfection was made in vitro with neuroblastoma murine cell line (Neuro‐2a) and in vivo with 21 years old mice. Both cells and mice were infected with different infectious doses (0.1; 1.0; 10 and 100 ID50) of the rabies virus. After 24 hours the infected cells and mice were transfected with polyclonal antibodies against rabies produced in equines (unpurified hyperimmune serum ‐ total antibody) using a cationic reagent, the lipofectamine 2000 previously diluted 1:20, for the negative transfection control, only minimum essential medium was used. Rabies is an infectious disease caused by a virus belonging to the family Lissavirus that affects all mammals including humans. Rabies transmission occurs by virus-containing saliva , through biting or licking of an injured tissue. The virus replicates at the site of inoculation until it reaches the nerve endings. At the neuromuscular junctions, the virus binds specifically to the nicotinic acetylcholine receptor. On the other hand, alkaloids have yielded interesting results in terms of drug development. The aim of this study was to develop a new mechanism for antiviral therapy against rabies based on the introduction by transfection with a cationic reagent (lipofectamine 2000) of antibodies into neuronal cells infected with the rabies virus. N2A cells were infected using 96-well plates and different viral concentrations (0.1, 1.0, 10 and 100 TCID50) of three lineages of rabies virus circulating in Brazil (dog, Desmodus rotundus and Eptesicus furinalis). Among the factors that establish the pre‐analytical variability, some studies report that hemolysis is the main interferential in several tests. However, no studies were found on immunofluorescence tests or that using cell cultures. The aim of this study was to evaluate the interference of different degrees of hemolysis serum samples in the evaluation of virus neutralizing antibodies (VNA) by rapid fluorescent focus inhibition test (RFFIT). The Fluorescent Antibody Virus Neutralization Test (FAVN) is used routinely in many laboratories for the reference measurement of rabies virus neutralizing antibodies (VNA) in serum of animals to confirm the efficacy of vaccination against rabies, which is required to authorize the transit of these animals in countries free of rabies. The World Organization for Animal Health (OIE) recommends a VNA title ≥ 0,5 IU/mL to ensure that the animal has immunity against the rabies virus.
